Sickle Cell Disease

A child inherits two copies of a mutated hemoglobin gene (HbS). When oxygen runs low, the HbS protein inside red blood cells clumps together and bends the cells into a stiff crescent (sickle) shape. These sickle cells jam in small blood vessels and break apart, starving tissue and triggering pain and organ damage.

These sickle cells jam in small blood vessels and break apart, starving tissue and triggering pain and organ damage.

Rush-hour traffic jam from a broken car

Imagine a highway where every car is supposed to be a flexible, squishy sedan that can squeeze through tight tunnels (normal hemoglobin A in soft red cells). One driver inherited a defective car blueprint and built a rigid, angular vehicle (hemoglobin S). When traffic slows down or the car gets cold, the rigid car's edges lock together with other rigid cars nearby, forming an immovable chain (polymerization). These stuck cars jam in the narrow tunnel exits (capillaries), backing up traffic and leaving towns downstream without supplies (vaso-blockage (occlusion) and tissue starved blood flow (ischemia)). The damaged cars break apart faster after a few days instead of lasting months, so the factory works overtime making replacements but can't keep up (hemolytic anemia and reticulocytosis). Meanwhile, the toll booth operator (spleen) who was supposed to pull broken cars off the highway gets overwhelmed, shuts down, and stops checking for counterfeit tickets (functional asplenia and loss of opsonized bacteria clearance).
